G-banding and Fluorescence In Situ Hybridization in Childhood Acute Myeloid Leukemia from South India

چکیده
Bone marrow samples obtained from the patients were cultured in RPMI 1640 (Gibco, Invitrogen, USA) medium that contained 15% fetal bovine serum for 24 and 48 hours at 37 C. After incubation, the cells were exposed to colcemid (0.10 μg/mL, Gibco) for 30 minutes, followed by hypotonic treatment (0.075 M KCl) Abstract Background: ence of recurrent cytogenetic abnormalities. Diagnostic clonal chromosomal abnormalities provide important prognostic information and are among the most important factors in predicting initial response to chemotherapy, duration of remission and overall survival. Methods: emia (AML). Results: The culture success rate was 94%, clonal chromosomal abnormalities constituted 62% and recurrent chromosomal abnormalities were 56%. In the favorable prognostic category, there were 51.6% of cases with t(8;21)(q22;q22), 16.1% had t(15;17)(q22;q21), and a total

G-banding and fluorescence in situ hybridization in childhood acute myeloid leukemia from South India.
BACKGROUND The current WHO classification of hematologic malignancies defines distinct entities of myeloid disorders based on the presence of recurrent cytogenetic abnormalities. Diagnostic clonal chromosomal abnormalities provide important prognostic information and are among the most important factors in predicting initial response to chemotherapy, duration of remission and overall survival. ...
متن کاملDetection of abl/bcr Fusion Gene in Patients Affected by Chronic Myeloid Leukaemia by Dual-Colour Interphase Fluorescence in situ Hybridisation
Conventional cytogenetic is the standard technique for detection of Philadelphia (Ph) chromosome in chronic myeloid leukemia (CML). Evaluation of abelson murine leukemia/breakpoint cluster region (abl/bcr) fusion using dual-colour fluorescence in situ hybridization (D-FISH) is an alternative approach allowing rapid and reliable detection of the disease. We employed the technique of interphase D...
متن کاملMolecular Cytogenetic Analysis of Gene Rearrangements in Childhood Acute Lymphoblastic Leukemia
The aims of this study were to estimate the incidences of BCR/ABL, MLL, TEL/AML1 rearrangements, and p16 deletions in childhood acute lymphoblastic leukemia (ALL), to identify new abnormalities, and to demonstrate the usefulness of interphase fluorescence in situ hybridization (FISH). We performed G-banding analysis and FISH using probes for BCR/ABL, MLL, TEL/AML1 rearrangements, and p16 deleti...
متن کاملPreferred co-localization of chromosome 8 and 21 in myeloid bone marrow cells detected by three dimensional molecular cytogenetics.
The impact of chromosome architecture in the formation of chromosome aberrations is a recent finding of interphase directed molecular cytogenetic studies. Also positive correlation of translocation frequencies and spatial proximity of chromosomes was described. Thus, disease specific chromosomal translocations could be due to tissue specific genomic organization. However, no three-dimensional i...
متن کاملComparison of I-FISH and G-banding for the detection of chromosomal abnormalities during the evolution of myelodysplastic syndrome.
Myelodysplastic syndrome (MDS) patients with a normal karyotype constitute a heterogeneous group from a biological standpoint and their outcome is often unpredictable. Interphase fluorescence in situ hybridization (I-FISH) studies could increase the rate of detection of abnormalities, but previous reports in the literature have been contradictory. We performed I-FISH and conventional karyotypin...
